Barenia M-75

Description

Pregabalin in Barenia M75 has approval from the United States Food and Drug Administration (FDA) for the treatment of neuropathic pain associated with diabetic peripheral neuropathy, spinal cord injury, and postherpetic neuralgia. Pregabalin is also approved by FDA for the treatment of fibromyalgia and as adjunctive therapy for partial-onset seizures in adults with epilepsy. Off-label uses include generalized anxiety disorder, social anxiety disorder, bipolar disorder, insomnia, and chronic pain conditions not otherwise approved by the FDA.

Methylcobalamin (MeCbl), the activated form of vitamin B12, has been used to treat some nutritional diseases and other diseases in clinic, such as Alzheimer’s disease and rheumatoid arthritis. As an auxiliary agent, it exerts neuronal protection by promoting regeneration of injured nerves and antagonizing glutamate-induced neurotoxicity.

MODE OF ACTION:

Pregabalin binds to presynaptic voltage-gated calcium channels at the alpha-2-delta subunit in central nervous system tissues. Binding of the alpha-2-delta subunit decreases the depolarization-induced influx of calcium into neurons and thus reduces the release of excitatory neurotransmitters. This action may account for the anticonvulsant and analgesic effects of pregabalin.

Methylcobalamine improved nerve conduction velocity; promoted injured nerve regeneration, recovering the neuromuscular functions in peripheral hyperalgesia and allodynia; MeCbl inhibited the ectopic spontaneous discharges from peripheral primary sensory neurons in neuropathic pain states. As a vitamin, MeCbl may be a potential candidate for treating peripheral neuropathy with good safety.
