Esomeprazole, Domperidone, Levosulpride

Esomeprazole is used to treat gastroesophageal reflux disease (Acid reflux) and indigestion by relieving the symptoms such as heartburn, stomach pain, or irritation. It also neutralizes the acid in the stomach and promotes easy passage of gas to reduce stomach discomfort.

It belongs to a group of medicines called proton pump inhibitors. It reduces the amount of acid your stomach makes which prevents further damage to the ulcer as it heals naturally. It may be given other medicines along with this medicine depending on what caused the ulcer.

Esomeprazole + Domperidone are a combination of two medicines: Domperidone and Esomeprazole. This combination is used to treat acidity and heartburn or gastroesophageal reflux disease (GERD); a condition where the acid in the stomach flows back up into the food pipe (esophagus). It is also used to treat gastric and duodenal ulcers. Domperidone helps to control vomiting by increasing the movement of the gut, allowing the food to move more easily through the stomach.

Levosulpiride is an enantiomer of sulpiride that exhibits anti-emetic, anti-dyspeptic, anti-psychotic and anti-depressant activities. Levosulpiride specifically blocks dopamine D2 receptors in the central nervous system (CNS) and gastrointestinal (GI) tract.

COMPOSITION:

Esomeprazole 40 mg Tab.

Esomeprazole 40 mg + Domperidone 30mg Cap.

Esomeprazole 40 mg + Levosulpride 75mg Cap.

MODE OF ACTION:

Esomeprazole exerts its stomach acid-suppressing effects by preventing the final step in gastric acid production by covalently binding to sulfhydryl groups of cysteines found on the (H+, K+)-ATPase enzyme at the secretory surface of gastric parietal cells. This effect leads to the inhibition of both basal and stimulated gastric acid secretion, irrespective of the stimulus.

Esomeprazole is a proton pump inhibitor (PPI) that works by reducing the amount of acid in the stomach which helps in the relief of acid-related indigestion and heartburn.

Domperidone is a prokinetic which works on the upper digestive tract to increase the movement of the stomach and intestines, allowing the food to move more easily through the stomach.

Levosulpiride is a sulpiride isomer that exerts its prokinetic action through a dual mechanism: 1) as a dopamine receptor antagonist and 2) as a serotonin 5HT receptor agonist, conferring this drug with a cholinergic effect. At a dosage of 25mg three times daily, levosulpiride accelerates gastric and gallbladder emptying.
